The relationship between the plant and rhizosphere microorganisms divides into either
positive interaction or negative interaction, depends on how the rhizosphere microorganism
affect on the plant. Some pathogens infect the plant and affect its defense mechanisms making
the plant more able to host the pathogen. Especially, Ralstonia solanacearum which is a harmful
pathogen that exploits any root wound or natural opening and infects the plant. Which case crop
misfortune in large numbers annually. This pathogen has the ability to infect many plants such as
Banana, tobacco, and tomato. It can transmit through soil, water, and hardware, and spreading to
the root cortex and parenchyma further invading to the xylem causing plant wilt. The aim of this
study was to enhance the plant defense system by creating a powerful bio-control agent that’s
based on rhizosphere organism. That will make the plant able to defend against the pathogen and
protect itself. In this study, 12 isolates were identified. followed with the inoculation of tomato
seeds and further greenhouse experiment. 7 isolates were shown positive effect and continued
with determining the rhizocompetence of these isolates and finalized with a second greenhouse
experiment followed with measuring the gene expression level of inducing systemic resistance
genes ; LoxA, Pin2, GluA and PR-1a of the plant which is dependent in controlling the synthesis
of JA and SA in order to elucidate the effect of bacterial inoculation on plant defense system
mediated by salicylic acid and jasmonic acid signaling pathway,and to determine which isolate
have the highest effect on enhancing the plant defense system. The results have shown that all
bacterial isolates succeeded in different rates in enhancing the plant defense system.

تنقسم العلاقة بين النبات والكائنات الحية المجهرية الموجوده بالتربة المحيطة بجذور النبات إلى تفاعل إيجابي
او تفاعل سلبي ، يعتمد على كيفية تأثير تلك الكائنات الحية الدقيقة على النبات. بعض مسببات الأمراض
تصيب النبات وتؤثر على آليات دفاعها مما يجعل النبات أكثر قدرة على ان تصبح عائلا لمسببات الأمراض.
على وجه الخصوص Ralstonia solanacearum ، الذي يعد مرض ضارًا يستغل وجود أي جرح فى
الجذر أو انفتاح طبيعي ويصيب النبات. التي تؤدى الى فقد المحاصيل بأعداد كبيرة سنويا. هذا المرض لديه
القدرة على إصابة العديد من النباتات مثل الموز والتبغ والطماطم. يمكن أن ينتقل عن طريق التربة والمياه
والأجهزة الخاصة بالنبات ، وينتشر إلى سطح الجذر و ينتشرإلى النسيج الداخلى للجذرمما يسبب ذبول النبات.
الهدف من هذه الدراسة هو تعزيز نظام الدفاع النباتي عن طريق إنشاء مادة حيوية قوية للتحكم معتمدة على
الكائنات المحيطه بالجذور. وهذا سيجعل النبات قادرًا على الدفاع ضد العوامل الممرضة وحماية نفسه. في هذه
الدراسة ، تم تحديد 12 عزلة. تتبعها تلقيح بذور الطماطم و تجربتها فى الصوبة. أظهرت 7 عزلات تأثيراً
إيجابياً و تم تحديد كفاءتها على الجذور هذه العزلات وانتهت بتجربة ثانية داخل الصوبة متبوعة بقياس مستوى
الجينات المسؤلة عن المقاومة فى النبات ؛ LoxA و Pin2 و GluA و PR-1a التى تعتمد في التحكم في
تخليق حمض الجاسمونيك و حمض الساليسليك من أجل توضيح تأثير التلقيح البكتيري على نظام دفاع النبات
بواسطة حمض الساليسيليك ومسار إشارات حمض الجاسمونيك. أظهرت النتائج أن جميع العزلات نجحت
بنتائج مختلفة فى التأثير على تعزيز نظام الدفاع عند النبات.
